Abstract
Objective:1) The purpose of this study was to observe the changes of ATGL gene and protein expression in C57BL/6J mice at different time points in endurance training;2) To explore the possible molecular mechanism of ATGL changes after intraperitoneal injection of c AMP activator Forskolin, PPAR-inhibitor T0070907.Methods:Experiment 1, Endurance training last for four weeks of training C57BL/6J mice, the gastrocnemius and soleus muscle of ATGL gene and protein expression was determined by real-time fluorescence quantitative PCR and Western blotting;Experiment 2, ATGL gene and protein expression in gastrocnemius and soleus muscle are determined respectively daily intraperitoneal injection of forskolin, T0070907 and DMSO after 4 weeks treadmill endurance trained mice.Results:1) ATGL gene and protein of the soleus muscle expression increased in endurance exercise (P<0.05) , and the ATGL m RNA expression in exercise group was significantly higher than the control group in fourth week (P<0.05) , the expression of ATGL protein was significantly higher than that of control group in the third, and in the fourth weeks (P<0.05) ;2) ATGL m RNA and protein expression in the gastrocnemius muscle increased during endurance exercise (P<0.05) , ATGL m RNA and protein expression in exercise group was significantly higher than those in the control group in the third week and the fourth week of endurance training (P<0.05) ;3) ATGL gene and protein expression of soleus and gastrocnemius muscle in forskolin group for 4 weeks endurance training were significantly higher than those of the control group (P<0.05) , ATGLm RNA and protein of T0070907 group in the gastrocnemius muscle was significantly lower than those of the control group (P<0.05) ;ATGL gene expression in forskolin group was significantly higher than that of gastrocnemius muscle after 4 weeks endurance training (P<0.05) .Conclusion:Endurance training can lead to the increasement of ATGL gene and protein expression soleus and gastrocnemius muscle in the C57BL/6J mouse, which the c AMP and PPAR-γ, signal molecule are involved in.
